About MT-1
MT-1 (Melanotan-I, also referred to as afamelanotide) is a synthetic linear analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), a neuropeptide derived from the proopiomelanocortin (POMC) precursor. Structurally it is [Nle4, D-Phe7]-alpha-MSH: the native sequence with a norleucine substitution at position 4 and a D-phenylalanine at position 7, modifications that increase metabolic stability and receptor binding affinity relative to native alpha-MSH.

Unlike the cyclic analogue MT-2, MT-1 retains the linear 13-residue backbone of alpha-MSH and shows a preference for the MC1R subtype. That selectivity profile is why laboratory investigators use the two as complementary tool compounds when dissecting which melanocortin receptor subtype drives an observed effect in a given model system.
